Advantages:

  • Reduction of irritation of the gastric mucosa
  • Great range of release rates possible
  • Predictable bioavailability
  • Minimized fluctuations in plasma concentration of drug
  • High dose strength administration
  • Reduces the esophageal residence time, compared with hard gelatin capsules

Disadvantages:

  • Hard to manufacture when compared to other available technologies
  • Somewhat pH dependent, which is not be an issue with OROS
  • Coating conditions may influence release rates
  • Not all tablet presses will successfully compress MUPS
  • Weight variation, poor hardness, and friability
  • Potential content segregation of coated pellets in the tableting blend
